To directly identify the source of newly formed cardiomyocytes during zebrafish heart regeneration, our group and others established a genetic strategy to lineage-trace cardiomyocytes in the adult fish, based on the cre/lox system widely used in the mouse [10,11]. Using this approach, both groups found that heart regeneration in zebrafish is driven by preexisting cardiomyocytes, rather than by progenitor cells.

Cardiac myocyte cell cycle control in development, disease, and regeneration.

Cardiac myocytes rapidly proliferate during fetal life but exit the cell cycle soon after birth in mammals. Although the extent to which adult cardiac myocytes are capable of cell cycle reentry is controversial and species-specific differences may exist, it appears that for the vast majority of adult cardiac myocytes the predominant form of growth postnatally is an increase in cell size (hypert...

Building and re-building the heart by cardiomyocyte proliferation.

The adult human heart does not regenerate significant amounts of lost tissue after injury. Rather than making new, functional muscle, human hearts are prone to scarring and hypertrophy, which can often lead to fatal arrhythmias and heart failure. The most-cited basis of this ineffective cardiac regeneration in mammals is the low proliferative capacity of adult cardiomyocytes. However, mammalian...

Postnatal telomere dysfunction induces cardiomyocyte cell-cycle arrest through p21 activation

The molecular mechanisms that drive mammalian cardiomyocytes out of the cell cycle soon after birth remain largely unknown. Here, we identify telomere dysfunction as a critical physiological signal for cardiomyocyte cell-cycle arrest. We show that telomerase activity and cardiomyocyte telomere length decrease sharply in wild-type mouse hearts after birth, resulting in cardiomyocytes with dysfun...

A microRNA-Hippo pathway that promotes cardiomyocyte proliferation and cardiac regeneration in mice.

In contrast to lower vertebrates, the mammalian heart has limited capacity to regenerate after injury in part due to ineffective reactivation of cardiomyocyte proliferation. We show that the microRNA cluster miR302-367 is important for cardiomyocyte proliferation during development and is sufficient to induce cardiomyocyte proliferation in the adult and promote cardiac regeneration. In mice, lo...

Single-cell transcriptome and epigenomic reprogramming of cardiomyocyte-derived cardiac progenitor cells

The molecular basis underlying the dedifferentiation of mammalian adult cardiomyocytes (ACMs) into myocyte-derived cardiac progenitor cells (mCPCs) during cardiac tissue regeneration is poorly understood. We present data integrating single-cell transcriptome and whole-genome DNA methylome analyses of mouse mCPCs to understand the epigenomic reprogramming governing their intrinsic cellular plast...
